Overview

ZG005 + gemcitabine + cisplatin is an experimental combination therapy being investigated for the treatment of advanced solid tumors, including but not limited to HER2-negative gastric or gastroesophageal junction adenocarcinoma, esophageal squamous cell carcinoma, and other unresectable, locally advanced, recurrent, or metastatic solid tumors. ZG005 is a bispecific antibody targeting both PD-1 (programmed cell death protein 1) and CD47, acting to reverse immune evasion by tumor cells both by promoting T-cell mediated immunity via PD-1 inhibition and by blocking the CD47-SIRPα pathway, which inhibits macrophage-mediated phagocytosis[4][5]. Gemcitabine is a nucleoside analog used in chemotherapy that inhibits DNA synthesis, and cisplatin is a platinum-based chemotherapeutic that crosslinks DNA, leading to apoptosis. Currently, the combination is under investigation through clinical trials and has not received regulatory approval for any indication[1].
