ZGGS15 is a recombinant humanized bispecific IgG4 monoclonal antibody that targets both LAG-3 (Lymphocyte-activation gene 3) and TIGIT (T cell immunoreceptor with immunoglobulin and ITIM domain). By binding to these immune checkpoint receptors, ZGGS15 blocks their interaction with respective ligands (MHC-II for LAG-3 and CD155 for TIGIT), thereby reversing immune suppression of T cells and NK cells. This leads to enhanced activation, proliferation, and cytokine release from T and NK cells, restoring their anti-tumor activity. Preclinical studies have shown that ZGGS15 has potent anti-tumor efficacy as monotherapy or in combination with PD-1 inhibitors such as nivolumab. It does not induce ADCC, CDC, or significant cytokine production, suggesting a favorable safety profile. The drug is being developed primarily for the treatment of advanced solid tumors[1][5][8].
