ZGGS18 is a bifunctional recombinant humanized antibody fusion protein that targets both vascular endothelial growth factor A (VEGF-A) and transforming growth factor beta (TGF-beta). It consists of an optimized bevacizumab-derived monoclonal antibody against VEGF-A fused to an engineered extracellular domain of the human TGF-beta receptor II. By simultaneously inhibiting VEGF-A and "capturing" TGF-beta isomers, ZGGS18 exerts antineoplastic effects through dual mechanisms—blocking tumor angiogenesis and reducing tumor metastasis. Additionally, it modulates the tumor microenvironment, potentially enhancing the efficacy of immune checkpoint inhibitors such as anti-PD-(L)1 antibodies. The drug is being developed primarily for advanced solid tumors and has shown synergistic effects in preclinical models when combined with immunotherapies[1][2][3][4][5][6].
