ZGN-1062

ZGN-1062 is a preclinical-stage methionine aminopeptidase 2 (MetAP2) inhibitor and a fumagillin derivative originally developed by Zafgen. It acts as a caloric restriction mimetic, reducing food intake and body weight. In preclinical models, ZGN-1062 has been shown to activate p53 signaling, leading to downstream induction of growth differentiation factor 15 (GDF15). It has been investigated for its potential to improve glycemic control, enhance insulin responsiveness in adipocytes, and extend healthspan and lifespan in aging models. Following Zafgen's merger with Chondrial Therapeutics to form Larimar Therapeutics in 2020, development of Zafgen's MetAP2 inhibitor pipeline, including ZGN-1062, was discontinued.
