ZGN-1258 is a second-generation, novel investigational small molecule drug developed as a potent and selective inhibitor of methionine aminopeptidase 2 (MetAP2) for the treatment of rare metabolic disorders, particularly Prader-Willi syndrome (PWS)[1][2][3][5]. Its mechanism involves inhibiting MetAP2, resulting in altered fat metabolism and suppression of hyperphagia (excessive hunger)[1][2][3]. In preclinical studies, ZGN-1258 demonstrated significant weight loss in obese mice by acting both on adipose tissue and neural hunger centers, with an expanded central action compared to earlier compounds in the same class[2][3]. However, toxicological studies in rodents revealed unexpected muscle tissue degeneration, which led to suspension of further clinical development and regulatory filings[1][2]. ZGN-1258 is not currently in trials due to these safety concerns.
