Drug intelligence / Profile preview
ZHER2-ABD-MMAF (also referred to as ZHER2-ABD-mcMMAF) is an experimental, preclinical-stage affibody-drug conjugate (AffiDC) designed for the targeted treatment of HER2-overexpressing malignancies, such as ovarian cancer. It consists of a human epidermal growth factor receptor 2 (HER2)-targeting affibody molecule (ZHER2:2891) fused to an albumin-binding domain (ABD) for in vivo half-life extension, and conjugated to the highly potent cytotoxic payload monomethyl auristatin F (MMAF) via a non-cleavable maleimidocaproyl (mc) linker. By binding to serum albumin, the ABD prolongs the circulation time of the conjugate. Upon binding to HER2 on cancer cells, the conjugate is internalized, and the MMAF payload is released to inhibit tubulin polymerization, leading to cell cycle arrest and apoptosis. Preclinical studies conducted by researchers at Uppsala University and KTH Royal Institute of Technology have demonstrated that ZHER2-ABD-MMAF exhibits strong anti-tumor efficacy and complete tumor regression in mouse models of HER2-positive ovarian cancer.
