Overview

ZHPV16E7384 is an affibody molecule specifically engineered to target the Human Papillomavirus type 16 (HPV16) E7 oncoprotein. It is being investigated as a therapeutic agent for HPV16-positive cervical cancer. The molecule is internalized into target cells via dynamin- and caveolin-1 mediated endocytosis. Once inside the cell, ZHPV16E7384 binds to the HPV16 E7 protein, thereby interrupting the interaction between E7 and the retinoblastoma protein (Rb). By preventing E7-mediated degradation of Rb, the drug restores Rb levels, which subsequently leads to the downregulation of phosphorylated Rb (pRb), E2F1, cyclin D1, and CDK4. This molecular cascade results in G1/S phase cell cycle arrest and the inhibition of tumor cell proliferation.
