ZI-MA4-1 is an investigational, allogeneic "off the shelf" T cell receptor-guided natural killer (TCR-NK) cell therapy developed by Zelluna Immunotherapy. It is the world’s first TCR-NK therapy targeting the cancer-testis antigen MAGE-A4, which is broadly expressed across various solid tumors. The product is engineered by transducing healthy donor-derived peripheral blood NK cells with a full CD3 complex and a CD8 coreceptor, enabling these NK cells to express an affinity-enhanced TCR specific for HLA class I-restricted MAGE-A4 peptide complexes. This dual mechanism combines innate NK cytotoxicity with targeted TCR-mediated tumor recognition, aiming to overcome tumor heterogeneity and immune escape mechanisms that limit current cell therapies. ZI-MA4-1 has demonstrated promising preclinical efficacy and safety profiles and is advancing toward phase I/II clinical trials for advanced solid tumors[2][3][5][6].
