ZI-PR-1 is a preclinical-stage T-cell receptor-engineered natural killer (TCR-NK) cell therapy being developed by Zelluna Immunotherapy for the treatment of solid tumors. The therapy targets the PRAME (PReferentially expressed Antigen in MElanoma) tumor antigen, which is frequently overexpressed in various cancers but has limited expression in healthy tissues. By engineering allogeneic natural killer cells from healthy donors to express specific TCRs, ZI-PR-1 combines the precise antigen recognition of T-cells with the innate anti-tumor activity and favorable safety profile of NK cells. This 'off-the-shelf' approach allows for large-scale manufacturing and on-demand administration, potentially overcoming the logistical and safety challenges associated with autologous CAR-T or TCR-T therapies.
