ZIC5-targeting siRNA is an experimental RNA interference (RNAi) therapeutic designed to specifically silence the expression of the Zic family member 5 (ZIC5) gene. ZIC5 is a member of the ZIC family of zinc finger transcription factors and has been identified as a key driver of cell survival and chemoresistance in pancreatic ductal adenocarcinoma (PDAC). The siRNA functions by inducing the sequence-specific degradation of ZIC5 mRNA, leading to a reduction in ZIC5 protein levels. This downregulation subsequently inhibits the expression of its direct downstream target, RIO kinase 3 (RIOK3), which is critical for the survival of pancreatic cancer cells. Preclinical studies have demonstrated that systemic administration of ZIC5-targeting siRNA can significantly reduce tumor volume and sensitize PDAC cells to gemcitabine treatment.
