ZIF-8@DOX@MnO₂@RTX is an experimental multifunctional nanoplatform developed for the targeted chemoimmunotherapy of B-cell lymphoma. The system utilizes a Zeolitic Imidazolate Framework-8 (ZIF-8) core—a zinc-based metal-organic framework—to encapsulate the cytotoxic agent doxorubicin (DOX). The nanoparticle is coated with manganese dioxide (MnO₂), which serves to modulate the tumor microenvironment, and is surface-modified with the anti-CD20 monoclonal antibody rituximab (RTX) for active targeting of lymphoma cells. This integrated approach is designed to enhance tumor-specific delivery and therapeutic efficacy while reducing systemic toxicity. Preclinical data indicates that the nanoplatform promotes apoptosis and inhibits tumor growth and invasion by downregulating early growth response 2 (EGR2) signaling and the anti-apoptotic protein BCL2.
