Overview

Zifanocycline (KBP-7072) is a novel semisynthetic third-generation aminomethylcycline antibiotic in clinical development for the treatment of drug-resistant bacterial infections. It acts by inhibiting the normal function of the bacterial ribosome through binding to the 30S ribosomal subunit, thereby blocking protein synthesis. Zifanocycline demonstrates broad-spectrum antibacterial activity against both Gram-positive and Gram-negative pathogens, including multidrug-resistant strains such as methicillin-resistant Staphylococcus aureus (MRSA), penicillin-nonsusceptible Streptococcus pneumoniae (PRSP), vancomycin-resistant Enterococcus faecium (VRE), and carbapenem-resistant Acinetobacter baumannii. The drug has shown potent in vitro activity even against isolates with common tetracycline resistance mechanisms like efflux pumps and ribosomal protection proteins. Both oral and intravenous formulations are being developed by KBP Biosciences[1][2][3][4][5][6][7].
