Drug intelligence / Profile preview
ziftomenib + fludarabine + cytarabine is an investigational combination chemotherapy regimen being evaluated for the treatment of pediatric and young adult patients with relapsed or refractory acute leukemias harboring KMT2A rearrangements, NUP98 rearrangements, or NPM1 mutations. The regimen combines ziftomenib (Komzifti), an oral small-molecule menin inhibitor, with the standard chemotherapy agents fludarabine and cytarabine (the FLA regimen). Ziftomenib works by blocking the interaction between menin and KMT2A (MLL) proteins, thereby disrupting the leukemogenic gene expression program. Fludarabine and cytarabine are nucleoside analogs that synergistically inhibit DNA synthesis and repair, with fludarabine enhancing the intracellular accumulation of cytarabine's active metabolite. This combination is currently being studied in a Phase 1 clinical trial (NCT06376162) sponsored by Blood Cancer United and Princess Maxima Center, with ziftomenib developed under a collaboration between Kura Oncology and Kyowa Kirin.
