Zigakibart is a first-in-class, humanized IgG4-kappa monoclonal antibody that targets and inhibits APRIL (A Proliferation-Inducing Ligand; also known as TNFSF13), a cytokine involved in the survival and maturation of B cells and the production of immunoglobulins. By blocking APRIL, zigakibart reduces pathogenic IgA antibody production implicated in the pathogenesis of IgA nephropathy (IgAN). Clinical studies have shown that zigakibart leads to significant reductions in proteinuria and stabilization of kidney function in patients with IgAN. The drug is administered subcutaneously every two weeks. Zigakibart has demonstrated an acceptable safety profile with most adverse events being mild or moderate infections[5][6][8].
