The Zika Virus Whole Virion Purified Inactivated Vaccine (also known as ZPIV or TAK-426) is a monovalent, formalin-inactivated biologic vaccine candidate designed to prevent Zika virus (ZIKV) infection and its associated neurological and developmental complications, including congenital microcephaly and Guillain-Barré syndrome. The vaccine is derived from the PRVABC59 strain of the Zika virus (Asian lineage), produced in Vero cells, and adjuvanted with aluminum hydroxide to enhance the immune response. Its mechanism of action involves the induction of high-titer neutralizing antibodies targeting the ZIKV envelope (E) protein, thereby preventing viral entry into host cells. Developed through a collaboration between the Walter Reed Army Institute of Research (WRAIR), Sanofi Pasteur, and Takeda, the vaccine has demonstrated safety and immunogenicity in Phase 1 clinical trials, although commercial development has fluctuated with the changing epidemiology of the virus.
