**ZIKV-117** is a potent human monoclonal antibody that neutralizes Zika virus (ZIKV) strains across African, Asian, and American lineages by binding a conserved quaternary epitope on the E glycoprotein dimers. It cross-links E protein monomers within and between dimers, preventing their rearrangement into fusogenic trimers required for viral entry in endosomes, while exhibiting no cross-reactivity with dengue virus (DENV) to avoid antibody-dependent enhancement. Developed from survivors of ZIKV infection, it has demonstrated efficacy in mouse models, reducing fetal infection, placental damage, and lethality in pregnant and non-pregnant animals when administered prophylactically or therapeutically, with structural studies confirming its mechanism at 6.2 Å resolution via cryo-EM.[1][3][7][8]
