Overview

Zimlovisertib (PF-06650833) is a potent, selective, and orally active small molecule inhibitor of interleukin-1 receptor-associated kinase 4 (IRAK4). Developed by Pfizer, it was designed to modulate inflammatory signaling pathways downstream of Toll-like receptors (TLRs) and interleukin-1 receptors (IL-1Rs). The drug was primarily investigated for the treatment of autoimmune and inflammatory conditions, specifically rheumatoid arthritis (in patients with an inadequate response to methotrexate) and hidradenitis suppurativa. Despite reaching Phase 2 clinical trials for these indications, Pfizer discontinued its development.
