Drug intelligence / Profile preview
Zirconium-89-labeled messenger RNA lipid nanoparticles (89Zr-labeled mRNA-LNPs) are radiolabeled nanoparticles developed by Okayama University and the University of Tokyo as a positron emission tomography (PET) imaging probe. Designed as a non-invasive tool for the in vivo pharmacokinetic and biodistribution evaluation of mRNA-based therapeutics, these nanoparticles are formulated with a lipid mixture (including ALC-0315, DSPC, cholesterol, ALC-0159, and DSPE-PEG(2000)-Azide) encapsulating EGFP mRNA. The nanoparticles are radiolabeled with zirconium-89 (89Zr) via click chemistry using a DFO-DBCO chelator. This diagnostic agent enables real-time, quantitative visualization of mRNA-LNP migration from the injection site to target tissues such as the lymph nodes, liver, and spleen, facilitating the preclinical optimization and clinical translation of mRNA delivery systems.
