Zirdafulven (LP-184) is an investigational small-molecule acylfulvene prodrug being developed by Lantern Pharma. It is designed as a precision oncology therapeutic that is selectively activated by the enzyme prostaglandin reductase 1 (PTGR1), which is frequently overexpressed in various solid tumors. Upon activation, zirdafulven induces potent DNA damage that preferentially targets cancer cells with deficiencies in the nucleotide excision repair (NER) pathway. This mechanism allows for the targeted treatment of tumors with high PTGR1 expression and DNA damage repair vulnerabilities. Zirdafulven is currently being evaluated in clinical trials for several high-unmet-need oncology indications, including advanced solid tumors, glioblastoma (GBM), triple-negative breast cancer (TNBC), and pancreatic cancer. The drug has received multiple FDA designations, including Fast Track for TNBC and GBM, and Orphan Drug Designation for malignant gliomas, pancreatic cancer, and atypical teratoid/rhabdoid tumor (ATRT).
