COF008, also known as ZK-H08, is an orally bioavailable, highly selective small molecule inhibitor of the RET (Rearranged during Transfection) tyrosine kinase. Developed by Lee's Pharmaceutical and its subsidiary ZKGZ Biotech (Zhaoke Oncology), the drug targets RET alterations, including fusions and mutations, which are known drivers in various cancers such as non-small cell lung cancer (NSCLC) and medullary thyroid cancer. By inhibiting the kinase activity of RET, COF008 blocks downstream signaling pathways that promote tumor cell proliferation and survival. It is designed to be potent against both wild-type RET and various drug-resistant RET mutants. As of recent pipeline updates, COF008 is undergoing Phase 1 clinical evaluation for the treatment of advanced solid tumors harboring RET genetic alterations.
