ZK2401 is a recombinant humanized bispecific antibody developed by Lee's Pharmaceutical for the treatment of advanced solid tumors. It is designed to simultaneously target Programmed Death-Ligand 1 (PD-L1) and 4-1BB (CD137). The molecule functions by blocking the immunosuppressive PD-1/PD-L1 checkpoint while providing a localized costimulatory signal through 4-1BB to activate tumor-infiltrating T cells. This dual-targeting approach aims to improve therapeutic efficacy and reduce the systemic toxicities often associated with systemic 4-1BB agonists. ZK2401 is currently undergoing Phase 1 clinical evaluation to assess its safety, tolerability, and preliminary efficacy in patients with solid tumors.
