Overview

**ZKA190** is a human monoclonal antibody that potently cross-neutralizes multiple lineages of Zika virus (ZIKV) strains by binding to an exposed epitope on domain III (DIII) of the viral envelope (E) protein, specifically the lateral ridge (LR) region and DI-DIII linker. It binds all 180 E protein copies on the virus surface, altering quaternary structure, inducing aggregation, and inhibiting post-attachment membrane fusion, with IC50 values of 0.004-0.05 nM. Developed from human donors, it demonstrates strong prophylactic and therapeutic efficacy in mouse models of lethal ZIKV infection, protecting against morbidity and mortality even when administered days post-infection, though viral escape mutants can emerge; a bispecific version (FIT-1) combining ZKA190 with ZKA185 prevents escape.[1][3][4]
