**ZKA78** is a human monoclonal antibody that targets the Zika virus (ZIKV) envelope (E) protein, specifically exhibiting neutralizing activity against ZIKV. It demonstrates potent inhibition of ZIKV E protein-mediated membrane fusion and neutralizes ZIKV African strains with an IC50 of 93 ng/ml. While effective against ZIKV in vitro, ZKA78 cross-reacts with dengue virus (DENV) and only partially neutralizes ZIKV infection, potentially promoting antibody-dependent enhancement (ADE) in vivo, as evidenced by severe symptoms and death in DENV-infected mice pretreated with ZKA78. It has been used as a positive control in studies assessing ZIKV neutralization in cell models like U937 cells.[3][4][7]
