ZKN-157 is a novel small molecule macrolide that functions as a Ribosome Modulating Agent (RMA). It is designed to exploit cancer ribosome heterogeneity by selectively inhibiting the translation of proteins upregulated by the MYC oncogene, particularly those involved in the ribosome and protein translation machinery. ZKN-157 has demonstrated selective antiproliferative activity in colorectal cancer (CRC) cell lines and patient-derived organoids, specifically those belonging to the Consensus Molecular Subtype 2 (CMS2), which is characterized by MYC-driven addiction to high protein translation. Preclinical studies indicate that ZKN-157 is effective as a single agent and exhibits synergistic effects when combined with clinically approved DNA intercalating agents. The compound was developed by Zikani Therapeutics (now part of Lytix Biopharma).
