Overview

ZL-1211 is a monoclonal antibody engineered to target Claudin 18.2 (CLDN18.2), a tight junction protein highly expressed in certain gastric and other gastrointestinal cancers. Unlike earlier CLDN18.2-targeting antibodies, ZL-1211 is designed to enhance antibody-dependent cellular cytotoxicity (ADCC), enabling it to induce robust antitumor activity even in tumors with low CLDN18.2 expression that may not be eligible for other therapies targeting this antigen[1][2][4]. The drug activates natural killer (NK) cells, leading to increased production of inflammatory cytokines such as IFNγ, TNFα, and IL6 within the tumor microenvironment, further promoting antitumor immunity[1][2]. Clinical trials have shown preliminary evidence of safety and antitumor activity in patients with advanced solid tumors including gastric cancer and pancreatic cancer[3][4]. ZL-1211 is being developed by Zai Biopharmaceutical.
