Overview

ZL-1218 is a recombinant humanized monoclonal antibody developed by Zai Lab that targets the chemokine receptor type 8 (CCR8), which is selectively expressed on tumor-associated regulatory T cells (Tregs). By binding to CCR8, ZL-1218 depletes these highly suppressive intratumoral Treg cells via antibody-dependent cellular cytotoxicity (ADCC), thereby modulating the tumor microenvironment and enhancing anticancer immune responses. The drug features an enhanced Fc region to optimize its effector function. It is being investigated as both a monotherapy and in combination with pembrolizumab for advanced solid tumors, with ongoing global Phase 1 clinical trials[4][5][7][10].
