ZL-1222 is a next-generation, PD-1-targeted interleukin-12 (IL-12) immunocytokine developed by Zai Lab for cancer immunotherapy. It is designed to leverage the potent anti-tumor activity of IL-12 while reducing the systemic toxicity typically associated with IL-12 therapies. By fusing a potency-reduced IL-12 cytokine to an anti-PD-1 single-chain antibody, ZL-1222 aims to deliver targeted immune activation within the tumor microenvironment. Preclinical studies have demonstrated that ZL-1222 induces strong interferon-gamma (IFNγ) production and tumor cell killing in both PD-1-sensitive and resistant models, with improved safety compared to wild-type IL-12 administration.
