ZL0420 is a potent and selective small molecule inhibitor of bromodomain-containing protein 4 (BRD4), an epigenetic reader protein. Developed by researchers at the University of Texas Medical Branch, ZL0420 is being investigated for the treatment of inflammatory bowel disease (IBD), including ulcerative colitis and Crohn's disease. The compound works by binding to the bromodomains of BRD4, thereby disrupting its interaction with acetylated histone lysine residues and preventing the stabilization of NFκB on the promoters of pro-inflammatory genes. This inhibition blocks the pathological activation of the BRD4-NFκB signaling pathway, leading to the suppression of key inflammatory cytokines such as TNFα, IL-6, IL-17A, and IL-8. In preclinical studies using mouse models of colitis, ZL0420 has demonstrated the ability to significantly reduce mucosal inflammation and is also being evaluated for its potential in preventing colitis-associated colorectal cancer (CAC).
