Overview

ZL0454 is a potent and selective small molecule inhibitor of Bromodomain-containing protein 4 (BRD4), an epigenetic reader protein. Developed by researchers at the University of Texas Medical Branch, ZL0454 is designed to treat inflammatory bowel diseases (IBD), such as ulcerative colitis and Crohn's disease, by disrupting the BRD4-NFκB signaling axis. BRD4 is required for the stabilization of NFκB on the promoters of inflammatory genes and for histone H3 Lys122 acetylation (H3K122ac). By inhibiting BRD4, ZL0454 suppresses the expression of key pro-inflammatory cytokines, including TNFα, IL-6, IL-17A, and IL-8. Preclinical studies in mouse models of colitis have shown that ZL0454 significantly reduces mucosal inflammation with a favorable safety profile compared to traditional NFκB inhibitors.
