ZL0513 is a potent and selective small molecule inhibitor of bromodomain-containing protein 4 (BRD4), an epigenetic reader involved in the regulation of inflammatory gene expression. Developed primarily for the treatment of inflammatory bowel disease (IBD), including ulcerative colitis and Crohn's disease, ZL0513 is designed to suppress mucosal inflammation and prevent the progression to chronic disease or colitis-associated colorectal cancer. To improve therapeutic efficacy and minimize systemic toxicity, ZL0513 has been formulated into PEGylated poly(lactic-co-glycolic acid) (PLGA) nanoparticles. This nano-encapsulation strategy facilitates targeted delivery to the inflamed colonic epithelium and provides sustained drug release. Preclinical studies in animal models of colitis have demonstrated that oral administration of nano-encapsulated ZL0513 significantly reduces colonic inflammation at lower doses compared to systemic administration.
