Overview

ZL0590 is a proprietary small molecule epigenetic modulator that acts as a potent and specific inhibitor of bromodomain-containing protein 4 (BRD4). Developed for the treatment of inflammatory bowel disease (IBD), including ulcerative colitis and Crohn's disease, ZL0590 works by disrupting the protein-protein interactions between BRD4 and acetylated histone lysine residues. This inhibition prevents the stabilization of NF-κB binding on the promoters of inflammatory genes and blocks the activation of RNA polymerase II, thereby suppressing the expression of key pro-inflammatory cytokines such as TNF-α, IL-6, IL-17A, and IL-1β. Preclinical studies in animal models of colitis have demonstrated that ZL0590 significantly reduces mucosal inflammation and restores tissue architecture, suggesting its potential as a therapeutic candidate for patients who are resistant to conventional anti-TNFα therapies.
