In clinical trials, such as the Phase I **UKHVC Spoke 003** study (NCT01922284), ZM96GPN is administered alongside **CN54ENV** (a plasmid encoding the HIV-1 Env protein) to prime the immune system, followed by booster vaccinations with viral vectors (e.g., Modified Vaccinia Ankara/MVA-C) and recombinant proteins (e.g., CN54rgp140). The vaccine facilitates the endogenous production and presentation of essential HIV-1 structural and regulatory antigens to stimulate robust cellular and humoral immune responses targeting the clade C virus, which is prevalent in sub-Saharan Africa.
