Overview

ZMA001 is a fully human monoclonal antibody (IgG1) developed by Zymedi for the treatment of pulmonary arterial hypertension (PAH). Its mechanism of action involves specifically blocking the infiltration of inflammation-inducing macrophages into the lungs by inhibiting lysyl-tRNA synthetase 1 (KARS1)-dependent migration of monocytes and macrophages. This approach targets a key pathological process in PAH—pulmonary vascular remodeling and inflammation—rather than simply alleviating symptoms through vasodilation. Preclinical studies demonstrated that ZMA001 reduces immune cell infiltration, lung fibrosis, and improves heart function in animal models, with superior efficacy compared to existing therapies such as sildenafil. The drug has shown synergistic effects when combined with standard PAH medications. Safety evaluations in animals revealed no adverse effects even at high doses. The first-in-human phase 1a clinical trial is ongoing to assess safety and tolerability in healthy adults.
