ZN-A-1041 + pertuzumab + trastuzumab is a combination therapy being investigated for the treatment of advanced HER2-positive solid tumors, including metastatic HER2-positive breast cancer and HER2-positive breast cancer with brain metastases[1][3]. ZN-A-1041 is an oral, brain-permeable, selective HER2-targeted tyrosine kinase inhibitor. Pertuzumab and trastuzumab are monoclonal antibodies targeting different domains of the HER2 receptor; pertuzumab inhibits HER2 dimerization, while trastuzumab blocks HER2 signaling and stimulates antitumor immune mechanisms, including complement activation and antibody-mediated cellular toxicity[2][4]. The combination aims for enhanced HER2 blockade by simultaneous inhibition of HER2 signaling at multiple levels (extracellular and intracellular), potential synergistic activity in tumor growth inhibition, and improved control of brain metastases[3]. Developed as a novel maintenance regimen for treatment-refractory patients, this regimen is currently being tested in early-phase (Phase 1) clinical trials.
