ZNF304 siRNA-DANP is an experimental RNA interference therapeutic consisting of a small interfering RNA (siRNA) targeting the transcription factor ZNF304, encapsulated within a dual assembly nanoparticle (DANP). The DANP delivery system features a chitosan-tripolyphosphate inner core and a polylactic acid (PLA) outer coating, designed for sustained release and prolonged gene silencing (up to two weeks from a single dose). ZNF304 is a Kruppel-associated box (KRAB)-zinc finger protein that transcriptionally regulates Integrin beta 1 (ITGB1). By silencing ZNF304, the drug reduces ITGB1 expression, thereby sensitizing ovarian cancer cells to anoikis (detachment-induced apoptosis), inhibiting migration, and reducing tumor growth and microvessel density. It is primarily being investigated for the treatment of high-grade serous ovarian carcinoma.
