Zocilurtatug pelitecan is an investigational first-in-class antibody-drug conjugate (ADC) targeting Delta-like ligand 3 (DLL3). It consists of a humanized anti-DLL3 monoclonal antibody conjugated to a novel camptothecin derivative (C24, a topoisomerase 1 inhibitor) via a cleavable tripeptide-based linker, with a drug-to-antibody ratio (DAR) of 8. The ADC utilizes the proprietary TMALIN® (Tumor Microenvironment Activable LINker-payload) platform technology developed by MediLink Therapeutics. The TMALIN® platform is designed to maintain payload stability in circulation while enabling efficient dual-mechanism (extracellular and intracellular) payload release within the tumor microenvironment. Zocilurtatug pelitecan is being developed globally by Zai Lab for extensive-stage small cell lung cancer (ES-SCLC) and potentially other DLL3-expressing neuroendocrine tumors. The drug has received FDA Orphan Drug and Fast Track Designations for small cell lung cancer.
