zocilurtatug pelitecan + tarlatamab

ZL-1310 + Imdelltra (zocilurtatug pelitecan + tarlatamab) is an investigational combination regimen being evaluated for the treatment of extensive-stage small cell lung cancer (ES-SCLC). The combination consists of two different modalities targeting delta-like ligand 3 (DLL3), a protein highly expressed on SCLC cells. Zocilurtatug pelitecan (formerly ZL-1310) is an antibody-drug conjugate (ADC) comprising a humanized anti-DLL3 monoclonal antibody conjugated to a novel camptothecin derivative (a topoisomerase 1 inhibitor) payload via a protease-cleavable linker. Imdelltra (tarlatamab) is a first-in-class bispecific T-cell engager (BiTE) that binds to both DLL3 on tumor cells and CD3 on T cells, inducing T-cell-mediated tumor lysis. By combining an ADC that delivers a cytotoxic payload directly to tumor cells with a T-cell engager that recruits the immune system, this regimen aims to provide synergistic antitumor efficacy in patients with SCLC.
