Zoe Biosciences is developing a high-affinity monoclonal antibody targeting plasminogen activator inhibitor-1 (PAI-1) for the treatment of age-related and cardiometabolic diseases. The lead candidate antibody binds to human PAI-1 with a dissociation constant (KD) of 0.2 nM and functions by converting active PAI-1 into an inactive substrate form. PAI-1 is a well-validated target in longevity research, as it is a known driver of cellular senescence and contributes to multiple hallmarks of aging. By inhibiting PAI-1, this biologic therapeutic aims to address conditions such as heart failure with preserved ejection fraction (HFpEF), obesity, sarcopenia, and general cardiometabolic aging. The program is currently in the preclinical stage of development.
