zolbetuximab

Zolbetuximab is a chimeric immunoglobulin G1 monoclonal antibody that targets Claudin 18.2 (CLDN18.2), a tight junction protein abnormally exposed on the surface of certain cancer cells, particularly in gastric and gastroesophageal junction (GEJ) adenocarcinomas. By binding to CLDN18.2, zolbetuximab triggers immune-mediated mechanisms—antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC)—leading to targeted destruction of CLDN18.2-positive tumor cells[3][4][6][8]. It is approved for use in combination with fluoropyrimidine- and platinum-based chemotherapy as first-line treatment for adults with locally advanced unresectable or metastatic HER2-negative gastric or GEJ adenocarcinoma whose tumors are CLDN18.2 positive[1][3][7]. Zolbetuximab was originally developed by Ganymed Pharmaceuticals.
