Drug intelligence / Profile preview
zoldonrasib + cetuximab + leucovorin + fluorouracil + oxaliplatin is an investigational combination regimen being evaluated for the treatment of gastrointestinal solid tumors, specifically KRAS G12D-mutant colorectal cancer. The regimen combines zoldonrasib (RMC-9805), an oral, mutant-selective covalent tri-complex inhibitor of the active GTP-bound state of KRAS G12D, with cetuximab, an anti-EGFR monoclonal antibody, and the mFOLFOX6 chemotherapy backbone (leucovorin, fluorouracil, and oxaliplatin). This combination is designed to achieve deeper and more durable inhibition of the RAS/MAPK pathway by simultaneously targeting mutant KRAS G12D and blocking EGFR-mediated feedback reactivation, while utilizing standard-of-care chemotherapy to enhance cytotoxic tumor cell death. The regimen is being investigated by Revolution Medicines in the Phase 1/2 RMC-GI-102 platform study (NCT06445062).
