Drug intelligence / Profile preview
zoldonrasib + pembrolizumab + platinum-based chemotherapy is an investigational combination regimen being developed by Revolution Medicines for the first-line treatment of patients with metastatic RAS G12D-mutant non-squamous non-small cell lung cancer (NSCLC). The regimen combines zoldonrasib (RMC-9805), an oral, G12D-selective covalent tri-complex RAS(ON) inhibitor, with pembrolizumab, an anti-PD-1 monoclonal antibody, and standard-of-care platinum-based doublet chemotherapy (typically pemetrexed combined with cisplatin or carboplatin). Zoldonrasib functions by binding to cyclophilin A to form a binary complex, which then selectively binds to and inhibits the active, GTP-bound state of the mutant KRAS G12D protein, preventing downstream oncogenic signaling. Pembrolizumab acts as an immune checkpoint inhibitor by blocking the interaction between PD-1 and its ligands, thereby restoring T-cell-mediated anti-tumor immune responses. The addition of platinum-based chemotherapy provides cytotoxic cell death, potentially releasing tumor antigens to synergize with the immunotherapy and targeted RAS inhibition. This combination is currently being evaluated in the global Phase 3 RASolve 308 clinical trial.
