This combination is in clinical trials in patients with MTAP-deleted and RAS mutant metastatic pancreatic cancer (PDAC) and non-small cell lung cancer (NSCLC). The dual inhibition approach targets both oncogenic KRAS G12D signaling (by zoldonrasib) and drives synthetic lethality in MTAP-deficient cells (by TNG462), aiming to maximize anti-tumor efficacy in genetically selected populations.
