A **combination drug** consisting of **zoledronic acid**, a third-generation nitrogen-containing bisphosphonate, and **recombinant human PH20 hyaluronidase**, an enzyme that temporarily degrades hyaluronan in the subcutaneous space. Zoledronic acid inhibits osteoclast-mediated bone resorption by binding to bone hydroxyapatite and disrupting osteoclast function, leading to increased osteoclast apoptosis. Recombinant human PH20 hyaluronidase increases tissue permeability by degrading hyaluronan, which can facilitate the subcutaneous administration and absorption of companion drugs. The combination may potentially allow subcutaneous delivery of zoledronic acid, improving convenience compared to standard intravenous administration, while maintaining efficacy for indications such as osteoporosis, bone metastases, and hypercalcemia of malignancy[1][4][5].
