Overview

Zorifertinib (AZD3759) is an orally administered, brain-penetrant small molecule inhibitor of the epidermal growth factor receptor (EGFR) tyrosine kinase. It was specifically designed to treat non-small cell lung cancer (NSCLC) with central nervous system (CNS) metastases harboring EGFR exon 19 deletion or exon 21 L858R substitution mutations. Zorifertinib effectively crosses the blood-brain barrier and inhibits both wild-type and mutant forms of EGFR at nanomolar concentrations. Clinical trials have demonstrated significant improvement in systemic and intracranial progression-free survival compared to first-generation EGFR-TKIs in patients with advanced NSCLC and CNS metastases[1][2][3][5][6][7].
