Shingrix is a non-live, recombinant subunit vaccine developed by GSK for the prevention of herpes zoster (shingles) and postherpetic neuralgia. It consists of the varicella-zoster virus (VZV) glycoprotein E (gE), which is the most abundant glycoprotein in VZV-infected cells and a primary target for T-cell responses, produced in Chinese Hamster Ovary (CHO) cells. The vaccine is formulated with the AS01B adjuvant system, a liposome-based formulation containing 3-O-desacyl-4'-monophosphoryl lipid A (MPL), a Toll-like receptor 4 (TLR4) agonist, and QS-21, a saponin extracted from the Quillaja saponaria tree. This adjuvant system is designed to enhance the magnitude and duration of the cellular and humoral immune response, particularly in older adults and immunocompromised individuals where immune senescence is a factor. It is administered as a two-dose intramuscular series.
