Zosurabalpin is an experimental *tethered macrocyclic peptide* (MCP) antibiotic developed by Roche and scientists from Harvard University. It targets *Acinetobacter baumannii* by inhibiting the LptB2FGC complex, disrupting lipopolysaccharide (LPS) transport, which leads to the accumulation of toxic intermediates and bacterial death. It is particularly effective against carbapenem-resistant strains and has shown potent activity in both in vitro and in vivo models. Zosurabalpin is being developed for the treatment of infections caused by carbapenem-resistant *Acinetobacter baumannii* (CRAB).
