Overview

ZP10000 (formerly ET3764) is an orally available cyclic peptide integrin inhibitor being developed by Zealand Pharma for the treatment of inflammatory bowel diseases. It acts as an antagonist of the gut-homing integrin alpha4beta7, disrupting lymphocyte trafficking to the intestinal mucosa and thereby reducing inflammation in the gastrointestinal tract. Originating from Encycle Therapeutics’ macrocyclic peptide platform and acquired by Zealand Pharma, the drug is positioned as a next-generation, oral alternative to infusion-only alpha4beta7 inhibitors such as vedolizumab, with a focus on gastrointestinal indications including ulcerative colitis and Crohn’s disease.[1][4][11][13][17]
