ZP10395 is a selective small-molecule inhibitor of ganglioside biosynthesis developed by Zacharon Pharmaceuticals. Gangliosides are a unique class of lipid-linked glycans that are frequently overexpressed in neural crest-derived tumors, such as melanoma, where they play a critical role in regulating growth factor signaling within lipid rafts. ZP10395 was identified using a novel molecular screening platform designed to find drug-like molecules that selectively reduce ganglioside levels without affecting other glycolipid classes, thereby potentially avoiding the dose-limiting toxicities associated with non-specific inhibitors. In preclinical studies, ZP10395 demonstrated 10-15 fold greater potency than existing non-specific inhibitors and significantly slowed tumor growth in mouse xenograft models of melanoma.
