ZRC-3277 + trastuzumab + docetaxel is a triple combination therapy used for the treatment of HER2-positive metastatic breast cancer. ZRC-3277 is a biosimilar of pertuzumab, a humanized monoclonal antibody that targets the extracellular dimerization domain (Subdomain II) of the human epidermal growth factor receptor 2 protein (HER2), thereby inhibiting HER2 heterodimerization and downstream signaling via MAP kinase and PI3K pathways, leading to cell growth arrest and apoptosis, and mediating antibody-dependent cell-mediated cytotoxicity (ADCC). Trastuzumab is a humanized monoclonal antibody that binds to the extracellular domain of HER2, leading to inhibition of proliferation and ADCC. Docetaxel is a taxane chemotherapy agent that stabilizes microtubules and prevents cell division. The combination is used as first-line therapy in metastatic HER2-positive breast cancer patients and is designed to maximize anti-tumor efficacy through blockade of multiple HER2 signaling mechanisms plus cytotoxic activity[1][3][5][7].
