ZS-1003 (also known as ZSSW-136) is a novel small-molecule topoisomerase I (TOPO1) inhibitor developed by Bio Genuine (Genuine Biotech). Discovered through an AI-Computer Assisted Drug Design (CADD) approach, the compound features a unique parent nucleus structure that differentiates it from traditional camptothecin-based drugs. This structural modification is designed to overcome drug resistance mechanisms associated with existing TOPO1 inhibitors, such as those found in irinotecan-resistant tumors. ZS-1003 is being developed for the treatment of various solid and malignant tumors, demonstrating significantly higher potency in resistant cancer cell lines compared to conventional payloads.
